Dementia studied in a model: Characterization of an animal model of Alzheimer's disease

Alzheimer's disease is the most common cause of senile dementia in the elderly population. Up to now, the clear mechanisms remain elusive due to huge complexity and a lack of knowledge about the pathology. There is no therapy available at present, however there is an impelling need to establish reliable screening methods and therapeutic tools.
The use of models for Alzheimer's disease and other challenging pathologies enables scientists to develop a better understanding about the etiology, to identify molecular markers and finally to develop therapeutic approaches.
This talk comprises the characterization of a highly suitable animal model for Alzheimer's disease. Within a longitudinal study, neurological and motoric skills, cognition, metabolism and biochemistry were investigated and analysed in order to provide a basis for preclinical research and validated the characterized animal model as appropriate for further studies.

MR-based Attenuation Correction in PET/MR-Systems – initial concept of BRUKER Biospin for small animal imaging

Nowadays combined PET/CT-Systems are commonly used to perform PET imaging with attenuation correction. However, more and more efforts are made to develop hybrid PET/MR-Systems with attenuation correction in order to benefit from the high tissue contrast of the MRI. Since MRI does not provide any knowledge about the attenuation properties of the tissue a different approach is needed to determine this information. This presentation will introduce an initial concept to derive an approximate attenuation map from the MRI data of the animal under investigation which then can be used to correct the attenuation effects. Also the impact of errors in the attenuation map will be addressed

Investigation of Macromolecular Protein Assemblies Using Solid-State NMR Spectroscopy Enhanced by DNP, Proton-Detection and Ultrafast Spinning

Abstract:
Many biologically interesting processes involve proteins or protein assemblies that are attached to or integrated into the cell membrane or other cellular/viral structures which are not dissolved in the cytoplasm. These insoluble proteins are difficult to study by X-ray crystallography or solution-state NMR.
In contrast, solid-state NMR spectroscopy has proven to be a successful technique to determine the structure of such proteins at atomic detail. One of the principal drawbacks of solid-state NMR is its inherently low sensitivity. In this talk, the two most promising approaches for sensitivity enhancement in solid-state NMR (dynamic nuclear polarization and proton detection) together with their applications to macromolecular protein assemblies are presented.
