Project update: Probing nociception in near physiological mice fMRI

Abstract:
Combining mouse genetics and functional magnetic resonance imaging (fMRI) provides a promising tool to unravel the molecular mechanisms of chronic pain. Probing murine nociception via the blood oxygenation level-dependent (BOLD) effect is still challenging due to methodological constraints. The aim of this presentation is to introduce an enabling methodology to permit the study of large-scale brain circuits in pain processing using near-physiological mouse fMRI. In contrast to established subcutaneous electrostimulation paradigms, the proposed method focuses on the reproducible application of brief, mild noxious heat stimuli (46 °C /14 sec) to the plantar hindpaw of anesthetized mice. This permits research into clinically relevant forms of chronic pain, such as hyperalgesia and allodynia. The method comprises further advances in technical, procedural and data processing tools for enhanced BOLD signal detection and a tight control of physiological parameters. The latter includes the development of a novel mouse cradle designed to maintain whole-body normothermia in anesthetized mice during fMRI in a way that reflects the thermal status of awake, resting mice. Applying this method to wildtype mice resulted in BOLD activation patterns closely resembling those observed in humans – a basic requirement for translational studies.

EMMOTRAC – embedded motion tracking for MR

Abstract: The topic of my talk will be our home-built optical tracking system for adaptive head motion correction in MRI. After a general introduction into adaptive motion correction, I am going to present results we have obtained in high-resolution anatomical imaging and functional imaging at 7T. Since September 2013, our group is funded by the German Ministry of Economics with the aim to start a business based upon our technology. Therefore, I will also give an overview about our goals during the grant period.

COSI-Measure: Open source multipurpose measurement system

Cost effective Open Source Imaging (COSI) aims to construct an affordable MRI scanner at low field using permanent magnets. In order to map the static magnetic field of the MR magnet, COSI-Measure was developed, which is an open source 3-axis multipurpose system for a total cost <3000€. Our system can be easily modified to be used for other applications such as electromagnetic field measurements, 3D printing, CNC machinery and many other scientific applications, that require submillimeter 3D movement.

Towards Quantitative Renal MR Blood Oximetry by Combined Monitoring of T2*, T2 and Blood Volume Fraction

Synopsis
Acute kidney injuries are often characterized by tissue oxygen hypoxia. T2*-mapping permits probing renal oxygenation but provides a surrogate rather than a quantitative measure of oxygen saturation (SO2). The link between pO2 and T2* is influenced by changes in blood volume fraction (BVf). Monitoring BVf in combination with recently developed quantitative BOLD approaches could permit unambiguous interpretation of renal T2*. To test the feasibility of this new approach we monitored renal T2*/T2 during baseline and short periods of venous occlusion. This was performed in the same animal under naïve conditions and again with USPIO to permit estimation of BVf and SO2.
